Understand the objective of the game. The goal of Manhunt is for the hunted to get from the starting point to the escape zone. The hunters have to find and "tag" the hunted before they get to the escape zone. The players both have their own bases. However, the hunted players have to reach their base.
The hunters start at their base and patrol around to keep an eye out for the hunted. Get at the minimum six players. Keep the amount of players even. Divide everyone into two equal teams and decide which team hides first.
Pick two captains of each team. Allow the captains to establish the bases of their teams - preferably close together. Play day or night. They both have their advantages and their disadvantages. Playing during the day grants more visibility and more time to play the game , but it may be hot and the streets will likely be busy.
Night will be cooler and less busy, as well as helping to hide the players in darkness, but your visibility is decreased, which increases the risk of people getting hurt or lost. If playing at night, make sure all players have flashlights and some form of a communication device, regardless of what team they're on.
Choose whether to play Captains Rule or Majority Rules. In Captains, if the captain of the hiding team gets to base untagged, then the hiding team wins. In Majority the more common and fun way , if the majority gets to base without being tagged, then that team wins. Even out the teams so that the game is fair. For example, if one of your players has asthma or is a slow runner, pair them up with an older or faster person.
Establish "out of bounds" areas. When establishing the rules of the game, make sure that everyone decides what areas are out-of-bounds, and if you are caught in these areas, you are automatically "tagged" and you have to go back to base.
Make sure players know that private property, cars, and abandoned or under-construction buildings are off-limits, since these could get you in trouble or be hazardous. Set a time limit. Come up with a time limit for the hiding team to hide and get to base, so you can get through a good number of games before you have to go in.

You could smelt cobblestone to make stone and dig down when they see you, then dig to the side, fill where you are with stone, and they'll think you're still digging. The possibilities are endless. As a Hunter, one of the worst things you can do is let the runner use portals. In order to beat the game, the runner has to visit both the Nether to collect blaze rods and the end to beat the dragon and thus win the game.
However, this can be difficult to stop as the runner is going to be bent on reaching the portals. Nether portals are hard to stop players from making, as lava pools are fairly common around the world, but if you use a water bucket you can turn the lava into obsidian and make it much harder for the runner to make the portal.
If you are in the nether you could use beds or TNT to break the portal or kill the runner when they come through. Don't just chase after the runner without a plan. You always want to make sure you maximize your chances of winning a fight by being aware of your surroundings and the runner. If the runner is smart they could have a plan or trap waiting to kill you, so be cautious when giving chase, and don't rush anything. There will always be another chance to kill the runner later, so don't take unnecessary risks.
It is always important to know when the right time to give a full on chase is. Know the Area: I'm not saying that you have to know where every nook and cranny is, but knowing enough to not get lost can be helpful. Don't Stay in the Same Spot: It's OK to stop and hide, but staying in the same spot for an extended period of time is just asking to be caught. Outmaneuver the Enemy: If the seekers figure out where you are, try to stealthily work your way around them and get away.
If they spot you and chase you, try weaving in and out of trees, you might get lucky and lose them. Avoid Crossing Open Areas: When moving from place to place, try to take the route with the most concealment, even if it's slower. Don't Wear Yourself Out: Heavy breathing will give you away, and you'll need your energy to outrun the seekers when they spot you. Think Like a Hider: Look where you would hide if you were in their shoes. Surround the Enemy: Work together with other seekers to try and corner the hider s.
Stay Together: Split into small groups or pairs to cover more ground, but avoid searching alone. If you do spot a hider, it's harder to outrun two people than it is to outrun one. Make Use of Trails: Move along trails and open spaces when possible, unlike a hider, speed, not concealment, is your greatest weapon. Watch for Movement: You're more likely to notice movement or sound than you are to actually see someone.
